Unexpected large changes in plasma antidiuretic hormone (ADH) levels are produced by manipulations of plasma osmolality. The objective of this study was to examine whether or not closed-loop osmotic regulation of ADH release from the pituitary gland could be achieved. Two intravenous infusions of hypertonic saline and dextrose (2-deoxy-D-glucose) were administered to three normal volunteers in a cross-over design. The stimuli for ADH release, administered by injection into the jugular vein, were a 10% glucose infusion at rates of 20, 30, and 40 ml/min. Plasma ADH concentration was measured in plasma samples collected every 5 min during the 10-min infusion. The two hypertonic infusions resulted in a sustained rapid increase in plasma ADH concentration, to an average of 147% above basal concentrations, but returned to basal values within 2 min after termination of the infusions. This indicates that acute alterations in plasma osmolality are reflected in increased pituitary secretion of ADH. The changes in ADH release were adequately described by a multiple regression model that included glucose infusion rate, duration of infusion, and an interaction term between glucose infusion rate and duration of infusion. A feedback model was used to simulate plasma ADH changes during the infusions. The model could reproduce the observed ADH increases during the infusions and the subsequent return to basal levels. In addition, it could be used to predict the plasma ADH concentrations during the infusions and to predict the effect of different infusion rates on ADH release.Cytoplasmic triglyceride droplets in isolated rat hepatocytes: relationship with the plasma membrane and lipogenic capacity of the cells.
